Abstract

Epidermal segments derived from flowering branches of Nicotiana plumbaginifolia were co-cultivated with a disarmed strain of Agrobacterium tumefaciens containing the co-integrate pTiBGSSSE∷pMONC9. Inoculated segments were subsequently grown on a medium for flower induction, and transgenic flowering shoots were recovered at a frequency of 40 percent. The transgenic plants obtained in vitro expressed the rbcS-8B-CAT gene in the appropriate organs. The flowers were fertile and set seeds which germinated with close to 100 percent frequency to give transgenic F1 progeny. Analysis of the kanamycin-resistant marker and CAT activity demonstrated that the transgenes were inherited as dominant Mendelian traits. With this technique it is possible to obtain transgenic plants with flowers and seeds six and eight weeks, respectively, after inoculation with Agrobacterium.
