Abstract

We have constructed an expression vector for the filamentous fungus Aspergillus nidulans using the well characterized amdS promoter system, and tested its potential as an expression system for foreign proteins using the Escherichia coli heat labile enterotoxin subunit B (LTB) gene. Co–transformation with a selectable plasmid was used to introduce an LTB construct into A. nidulans and transformants containing multiple copies of the construct were identified. DNA and mRNA coding for LTB were detected in transformants, and mRNA levels were inducible by acetate via the amdI9 mutation in the expression vector. Mature LTB was detected in cellular extracts but not in the medium, and expression levels correlated with the number of LTB genes.
